Design Aspects of a Computer Simulation Study for Assessing Uncertainty in Human Lifetime Toxicokinetic Models

Abstract

The general paradigm for risk assessment of exposures to toxic agents in human environment is the identification and characterization of hazard, assessment of exposure and characterization of risk. Performed in practice risk assessment is addressing particularly the outcome of integrating the data available from epidemiology, long-term mortality and morbidity studies and mechanistic research with information on the type and extent of exposure, as well as statistical analysis properly used. Various technical and non-technical aspects of the design process of the Monte Carlo simulation study will be reported and discussed. Finally, a Monte Carlo computer simulation study was designed in order to examine in detail the influences of various sources of uncertainty and their potential implications on the risk estimates from the Boehringer cohort data is presented.
