The Dbf4–Cdc7 kinase promotes S phase by alleviating an inhibitory activity in Mcm4

Abstract

Dual function of Dbf4–Cdc7 kinase Two cell-cycle-regulated kinase pathways regulate the timing of DNA replication initiation. How one of these, the Dbf4–Cdc7 kinase (DDK), affects replication has been unclear. Yi-Jun Sheu and Bruce Stillman now show that a subunit of the replicative helicase, Mcm4, is the target of DDK. The N-terminal domain of Mcm4 possesses an inhibitory activity that is relieved by DDK phosphorylation. In addition, DDK activity also seems to have a second function in regulating the S-phase checkpoint.
