Investigation of Linkage and Association‐Issues on Study Design

Abstract

For the identification and description of susceptibility genes for complex diseases using genetic markers there are two basic strategies: genome scans or the investigation of candidate genes. The designs can be quite different for the two approaches and decide, more than in general epidemiology, the methods that can be applied subsequently. Study design, recruitment and sample size are discussed further in the context of the Transmission/Disequilibrium Test, a family‐based simultaneous test for linkage and association. Deux approches sont utilisées, pour identifier des génes, de susceptibilitédes maladies complexes à I' aide de marquers génétiques: la recherche systématique sur tout le genome ou l' étude de génes candidate, les protocoles d'étude sont trés différents, dans les deux cas et cela détermine, plus que normalment en I'épidémiologie, les méthodes qui seront ensuite appliquées. Nous discutions ici du protocole de I' étude, du type et de la taille d'échantillon utilisé dans le context du Transmission/Disequilibrium Test, test simultané de liaiosn et d'association.
